- Research Topic: Epigenomic study of -amyloid formation resulting in neuronal dysfunction and loss Research Deatils: There exists a general recognition of the fact that a central event in the development of Alzheimers disease (AD) is the abnormal processing of neuronal cell membrane amyloid precursor protein followed by deposition of -amyloid protein in the form of amyloid plaques responsible for the activation of microglia leading to the production of toxic and inflammatory mediators like hydrogen peroxide, nitric oxide and cytokines. The present study was addressed to explore the interrelationship between Apoptosis antagonizing transcription factor (AATF), Prostate apoptosis response-4 (PAR-4) and Liver X receptor- (LXR-) in the generation of Amyloid leading to ROS production followed by neuronal apoptosis. Research Techniques: Expression levels of genes including AATF, PAR4, and LXR- were compared using real-time quantitative reverse transcription polymerase chain reaction, western blotting, and reporter assays. Moreover, the ability of LXR- to induce ROS production followed by neuronal apoptosis was verified by using anti-A antibody and various ROS chelators using Flow cytometry.
